Terumbu karang adalah sekumpulan hewan karang yang bersimbiosis dengan sejenis tumbuhan alga yang disebut zooxanthellae. Zooxanthellae merupakan kelompok dinoflagellata fototropik yang umumnya terdapat sebagai endosimbion pada beberapa invertebrata laut. Penelitian ini dilakukan pada bulan April 2017 yang berlokasi di pulau Gili Labak Kecamatan Talanggo Sumenep Madura, dengan Tujuan penelitian 1. Mengetahui kepadatan zooxanthellae pada karang sehat, bleaching dan penyakit white band disease (WBD). 2. Mengetahui indeks mitotik zooxanthellae pada karang sehat, bleaching dan peyakit white band disease (WBD). Kepadatan adalah jumlah sel yang berada pada jaringan inang yang diukur dengan satuan sel/cm 2 . Sedangkan indeks mitotik presentase adalah laju pembelahan sel. Sel zooxanthellae dipisahkan dari jaringan karang dengan cara disemprotkan menggunakan air laut yang steril. Tingkat kepadatan zooxanthellae kemudian dihitung menggunakan rumus APHA. Hasil yang diperoleh kepadatan zooxanthellae pada karang sehat memiliki nilai rata-rata sebesar 16040000, karang berpenyakit jenis Bleaching nilai rata-rata sebesar 1080000 dan karang berpenyakit jenis White Band Disease (WBD) memiliki nilai rata-rata sebesar 800000. Sedangkan nilai indeks mitotik pada karang sehat mendapatkan nilai rata-rata 15,33%, Bleaching 25,26% dan WBD 19,5%. Dari hasil uji korelasi hubungan kepadatan dan indeks mitotik zooxanthellae pada karang sehat memiliki hubungan 0,156 (sanggat renda), karang bleaching 0,4773 (sedang) dan karang white band disease 0,7018 (tergolong kuat).

Zoxanthellae cell separated from the coral network with sprayed using sea water sterile. The level of zoxanthellae density then calculating use the formula APHA, the result which obtained zooxanthellae on healthy coral has the average vawe of 16040000, coral disease which kind of bleaching with the average vawe 1080000 and coral disease which kind of white band disease (WBD) has the average vawe 800000. While the index mitotic vawe on healthy coral get the average vawe 15,33%, Bleavhing 25,6% and WBD 19,5%. From theresult of correlation test of connection the density and zooxanthellae mitotic index of healthy coral which have relationship 0,156 very low, bleaching coral 0,4773the average and white band disease coral with pertained 0,7018 powerful.
